Specialist – Bluestone Lab
Research in the Bluestone lab is broadly focused on understanding mechanisms regulating T cell activation. Our work has centered on altering the positive and negative co-stimulatory signals that are delivered in conjunction with signals from the T cell receptor during T cell activation. By manipulating positive co-stimulatory ligands, such as B7-1 or B7-2, or negative regulatory receptors, such as CTLA-4 or PD-1, we revealed new mechanisms to promote immunotolerance. In addition, we are studying an immunosuppressive population of T cells known as Tregs. Tregs are essential for preventing most forms of autoimmunity and we are developing strategies to utilize these cells to treat Type 1 Diabetes and other autoimmune diseases.
The breakdown of tolerance has been attributed to an imbalance of effector function and immune regulation, specifically defective regulation due to defects in the T regulatory cells (Treg) subset. Thus, multiple efforts have been forged to re-instate that balance in setting such as autoimmune disease and organ transplantation or disrupt it as a means to promote anti-tumor immunity. Recent investigations have focused on Treg instability in the autoimmune and cancer settings, and targeting of the FOXP3 pathway to selectively enhance Treg function. We have also focused attention on novel approaches to understanding FOXP3 activity and delivering specific signals to Tregs to promote Treg stability and function, including the use of novel IL-2 and anti-IL-2 approaches. Finally, we have initiated early clinical trials translating the insights gained from mouse studies to deliver Tregs and IL-2 therapeutically to promote rebalancing of effector and Treg function in autoimmunity and transplantation.
Your purpose in this position is to aid the clinical manufacturing team in performing assays and presenting data for review to lab managers. Once trained on the appropriate standard operating procedures (SOPs), you will be scheduled to perform assays and review data with your supervisor. You will also work on aiding in the development of new assays on high dimensional analysis platforms for immunophenotyping. You will also have the opportunity to work and learn on new instruments and analysis software packages in the Flow Cytometry Core. You will perform general assays, QC/QA following documented SOPs, and generate new SOPs and protocols if needed. Attendance at work is of the utmost importance and is directly related to job performance as the broad base of lab members rely and depend on your timely, predictable, and scheduled support. You will also participate in research, development, and testing of new instruments, reagents, and techniques, and perform other duties as assigned.
• Specialists appointed at the junior rank must possess a baccalaureate degree (or equivalent
degree) in microbiology/immunology or related field or at least 4 years of research experience in a laboratory environment.
• Specialists appointed at the Assistant or Associate rank must possess at minimum a master’s degree (or equivalent degree) in microbiology/immunology or related field or at least five years of experience in a laboratory environment.
• Specialists appointed at the Full rank must possess a terminal degree (or equivalent degree) in microbiology/immunology or related field or at least ten years of experience in a laboratory environment.
• Demonstrated ability to operate various flow cytometers such as BD FACSCalibur, FACSAria platform, and BD LSRII/Fortessa/X20 platform.
• Demonstrated knowledge of analysis software systems such as CellQuest, FACSDiVA, FlowJo flow cytometry software.
• Knowledge of safety procedures in a laboratory environment.
• Ability to perform calculations and statistics, understanding complex laboratory
protocols, methods and interpreting results.
• Excellent organizational, record keeping and time management skills. Must be flexible and able
to prioritize tasks. Careful attention to details.
• Excellent interpersonal, verbal and written communication skills. Ability to work with a
diverse group of people and interact with all levels; work independently and within a team environment.
• Proficiency with MS operations, including advanced skills in Word, PowerPoint, and Excel.
• Demonstrated ability to work independently to a high standard of accuracy and ability to
follow and operate in compliance with standard operating procedures.
• Demonstrated experience and ability to share knowledge and train users and other staff.
• Extensive expertise with flow and/or mass cytometry.
• Experience with and a mindset for teaching and supporting researchers with microscopy and flow cytometry
• Previous experience working in a research laboratory in the areas of cell and
molecular biology, genetics, and bioinformatics.
• Previous IT/system administrator experience with various operating systems, network
management, web design, programming.
• CCy certification would be preferred.
